Regulation of β-Disaccharide Accumulation by β-Glucosidase Inhibitors to Enhance Cellulase Production in Trichoderma reesei

چکیده

Trichoderma reesei is a high-yield producer of cellulase for applications in lignocellulosic biomass conversion, but its production requires induction. A mixture glucose and β-disaccharide has been demonstrated to achieve high-level production. However, as inducers, β-disaccharides are prone be hydrolyzed by β-glucosidase (BGL) during fermentation, therefore need supplemented through feeding overcome this problem. Here, miglitol, an α-glucosidase inhibitor, was investigated BGL exhibited IC50 value 2.93 μg/mL. The titer more than two-fold when miglitol added the fermentation medium T. reesei. This method similar prokaryotic expression system using unmetabolized isopropyl-β-D-thiogalactopyranoside (IPTG) inducer instead lactose continuously induce gene expression. activity not enhanced with inhibition or cellulose used inducer, which that transglycosidase important inducible cellulose. novel demonstrates potential stimulating provides promising protein

Cellulase Production by Trichoderma reesei using Sugar Beet Pulp

Cellulase production by the fungus Trichoderma reesei was studied using sugar beet pulp (SBP) as a substrate. The subculture medium was a salt solution consisting of KH2PO4, CaCl2, etc. Fungal cells were sub-cultured in an orbital shaker (180 rpm) at 30°C for 1-2 generations (two days for each generation) and were then used as an inoculum. Exponential cells were inoculated into a medium contain...

Production of Cellulase-free Xylanase by Trichoderma Reesei Saf3

A xylanase producing fungi has been isolated from soil and identified as Trichoderma reesei SAF3. Maximum growth of the organism was found at 48 h under submerged condition in xylan containing enriched medium, whereas highest enzyme production (4.75U/mL) was recorded at 72 h. No detectable cellulase activity was noted during whole cultivation period. The partially purified enzyme hydrolyzed xyl...

Increased Production and Activity of Cellulase Enzyme of Trichoderma reesei by Using Gibberellin Hormone

Cellulolytic complex are enzymes capable of hydrolyzing cellulose. Due to rapid growth in population and industrialization, most countries are required to produce more fuel. Production of bioethanol from lignocellulosic biomass is very challenging due to environmental pollution by fossil fuels. Cellulases play a significant role in biotechnological processes. The cost of production of cellulase...

Improvements in Glucose Sensitivity and Stability of Trichoderma reesei β-Glucosidase Using Site-Directed Mutagenesis

Glucose sensitivity and pH and thermal stabilities of Trichoderma reesei Cel1A (Bgl II) were improved by site-directed mutagenesis of only two amino acid residues (L167W or P172L) at the entrance of the active site. The Cel1A mutant showed high glucose tolerance (50% of inhibitory concentration = 650 mM), glucose stimulation (2.0 fold at 50 mM glucose), and enhanced specific activity (2.4-fold)...
